      Hi all. It’s been years since I’ve posted as I’ve been stage 3 since 1998. Now stage 4, lung nodule removed in right lung last year, now new tumors in right and left lungs. Did 3 months of yervoy and tumors spread in both lungs. Started zelboraf and after 8 days stopped due to severe rash. A week of steroids then half dose of zelboraf for a week. AST (liver functions) test went from 25 to 382 so now half of that dose (1 pill twice a day) and still have the rash. I have a pet scan next week and my question is, if liver function test still high and not much improvement, what is next?

          Had a similar experience, I will post the link here. I take Aleve & Prednisone both to stay on Zel and control pain& rash. Then can only go 2 wks or so and need a 1 wk break.

                Sandy – sorry to hear about the troubles with Z.  I did Z last May-Sept and Yervoy Sept-Nov (with surgery in Oct).  They weren't able to remove a lymph node in my intestine area and scans Dec '12 showed growth.  Started PD1 trial Jan 21st.  Scans right before did show shrinkage (So maybe delayed response?).  Have scans on PD1 coming up Apr 12th.  Have you talked about getting in a anti-PD1 trial?

                      In addition to anti-PD1, the combination of a BRAF inhbitor (dabrafenib) and a MEK inhibitor (trametinib) seems to work in more patients and with few side effects than does either one alone. Both drugs are in the FDA approval process and may be publically available soon.

                            I am in the process of investigating trials and will look closely at this one… have heard alot about it.  My liver functions have gone down to almost normal so oncologist wants to put me on 1 pill a day and try to increase gradually.  Last PET scan showed really good improvement for being on Z for such a short time.  Will try that for a while and monitor scans.  Thanks for the reply!
