      I started the combo today. I took the mekinist with the tafinlar this morning.  I am wondering if taking it with the evening dose would be easier..maybe sleep through side effects?  I have been very tired today..a bit shaky…slight headache and my face flushed. No fever yet… Has anyone had a red face or noticed that the timing of taking mekinist made a difference?  I read the side effects that are possible and just need some input.  Thank you!

          My husband started the combo three weeks ago, he takes the Tafinlar in the morning and 12 hours later he takes the Tafinlar + Mekinist.  It seems to work well that way.  He doesn't eat two hours before or one hour after any of the medications, as recommended.  His main symptom is a slight low grade fever and he has not had that for the past few days.  He is feeling great and no other symptoms noted!  He just had some blood work done and his labs are coming out fantastic so far.  He goes back in August for another scan.  Keeping our fingers crossed, but he is feeling better than he has in months! 🙂  We are very hopeful!  Hope it goes alright for you too!  We will be praying for  you as well.  Keep us posted 🙂  By the way we take his vitals every day and write down any symptoms he has had for that day.  Keeping a diary will help when visiting his Dr.  ( I am an RN, so I am a little obsessive with vitals and administration )

                  Sorry,  am new to all of this. HOw does the Tafinlar + Mekinist compare to Ipi + Nivo ????

                    I am not sure how it compares since we have not been on any other drug or treatment.  But I know if you have BRAF positive melanoma, this is a wonderful combo 🙂

                            I've been on the combo 23 months now and am tolerating it really well. I did hit the wall with side effects at the 5-6 week mark – dehydration, mouth ulcers, really bad leg/ joint pain , temps – you name it I had it!  – I had a good weeks break off meds & then resumed on 50mg dose of Dabrafenib & a slow weaning off prednisone . Still on same reduced dose of Mek & dabrafenib now. I exercise a lot & live a full & healthy life. I do get the odd headache still but nothing  paracetamol doesn't fix. My scans to date have shown NED & I'm hoping that continues. So hang in there – there's light at the end of the tunnel!

                             Re your flushed face – I found my skin went red/  speckley on my legs & red/ hot on soles of my feet & hands at times. The side effects seemed to be short lived & even cyclical for me. 

                            I take the combo at 5.30am & dabrafenib 5.30pm – I drink 3 big glasses of water & make sure I follow the time schedule for empty stomach.

                            I hope this helps – I know it's hard when you first start out – you just don't know what's ahead . Take care & all the best.
